Position Description

A Research Specialist position is available in the MechanoBiology Lab within the Department of Orthopaedic Surgery, located at the University of Pittsburgh School of Medicine. The incumbent must have solid knowledge in cell biology/molecular biology and also excellent skills in basic cell and molecular techniques.

Specifically, the following technical skills are highly desirable:
- Cell culture, cell screening, cell transfection, mouse primary cells isolation and culture, luciferase reporter assay, flow cytometry, Immunoprecipitation, and fluorescence microscopy.
- PCR, RT-PCR and real-time PCR techniques and primer design, DNA and RNA isolation and electrophoresis, Plasmid DNA construction, and expression of cloned genes in E. coli and mammalian cells.
- Southern Blotting, Northern Blotting, and in-situ hybridization.
- Protein electrophoresis (SDS-PAGE), Western Blotting, Silver Staining, Gel-shift Assay, or Electrophoretic Mobility Shift Assay (EMSA).
- Immunohistochemical assay, radioimmunoassay, and ELISA.
- Virus preparation, Virus copy number titration, and Virus infection of mammalian cells and mice.
- Transgenic, knock out, and normal mice maintenance and genotyping.
- Withdrawal of blood in animals, preparation of platelet-rich plasma (PRP), and analysis of blood components.
- Animal handling and experiments.

The incumbent will work on tendon/ligament mechanobiology and repair/regeneration of injured tendons and tendon-bone junctions using innovative tissue engineering approaches. He/she may also work on the projects on tendon stem cell mechanobiology and platelet biology related to the mechanisms of tendon/ligament injuries and repair of injured tissues.

A Master’s degree in biology, biochemistry, or a related life science field is required.
